Isolation, Properties and Kinetics of Growth of a Thermophilic Bacillus
نویسندگان
چکیده
A strictly aerobic, thermophilic, spore-forming bacteria was isolated from a soil sample collected in Campos dos Goytacazes City, Rio de Janeiro, Brazil. The cells of this organism were Gram-positive, catalase positive, actively motile, 1.2 μm wide and 5.3 μm long. Growth occurred at pH values ranging from 6.5 to 9.0, and optimum growth occurred at about pH 7.0. The optimum temperature for growth was around 55oC, and the upper temperature limit for growth was around 70oC. Yeast extract stimulated growth on glucose and was obligately required at supraoptimal temperatures (65oC). The results of 16S rRNA sequence comparisons indicated that the isolate was closely related to Bacillus caldoxylolyticus and Bacillus sp strain AK1, and these three organisms exhibited levels of ribossomal DNA sequence homology of 94%.

Prototrophic thermophilic bacillus: isolation, properties, and kinetics of growth.
A thermophilic bacillus (minimal growth temperature 41 C, optimal 55 to 58 C, and maximal 65 C) was isolated from a manure pile. It is very similar to Bacillus stearothermophilus, but it differs in its inability to hydrolyze starch. The thermophilic isolate is a prototroph which grows in a minimal medium consisting of glucose, ammonium salt, phosphate buffer, and inorganic salts. At all tempera...

The goal of this research was to isolate and identify the thermostable alkaline protease producing bacteria among several native Iranian microorganisms. At the end of screening program, a Bacillus subtilis BP-36 strain producing thermophilic alkaline protease was isolated from a hot spring in Ardebil province. The target enzyme was purified using a one-step Aqueous two-phase systems (ATPS) prot...
متن کاملIsolation and Characterization of Thermophilic Alkaline Proteases Resistant to Sodium Dodecyl Sulfate and Ethylene Diamine Tetraacetic Acid from Bacillus sp. GUS1
Thermophilic Bacillus sp. GUS1, isolated from a soil sample obtained from citrus garden, produced at least three proteases as detected by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and zymogram analysis. The enzymes were stable in the alkaline pH range (8.0-12.0), with the optimum temperature and pH range of the proteases being 70ºC and 6.0-12.0, respectively. All th...
متن کاملStudies on a thermophilic bacillus: its isolation, properties, and temperature coefficient of growth.
A thermophilic bacillus with minimal, optimal, and maximal growth temperatures of 40, 64.5, and 72 C, respectively, was isolated from soil. Biochemical and morphological studies place the isolate in group 1 of the classification of Walker and Wolf. After adaption to nitrate broth, the temperature coefficient for growth was found to be 20,400 cal/mol. When the temperature coefficient for growth ...
متن کاملIsolation and Partial Characterization of a Bacterial Thermostable Polymethyl Galacturonase from a Newly Isolated Bacillus sp. strain BR1390
Background: Pectinases are pectin degrading class of enzymes including polygalacturonase (PG), polymethyl galacturonase (PMG), pectate lyase (PEL), and pectin esterase (PE) that are commonly used in processes involving the degradation of plant materials, such as speeding up the extraction of fruit juices. Objectives: A highly methylated pectin degrading bacterium from soil covered with fruit wa...
